The 25-year-old striker had to be substituted as la Roja suffered a 1-0 loss to France in an international friendly in Paris on Thursday night.
And Costa, who was part of the Spain squad that flopped at the World Cup, is set to return to Chelsea to receive treatment on his hamstring injury.
A statement from the Spanish Football Federation said: “Diego Costa is not available for selection because of an injury to the hamstring muscle in his left thigh. All imaging tests will be sent to Chelsea.”
Earlier on Friday, Costa revealed that he has taken encouragement that he is improving as a player following an impressive start to his Chelsea career.
The Spain international has scored four goals in his first three Premier League games, helping the west London side take maximum points to go top of the table.
“I really want to score with Spain,” Costa told Marca.
“Our game is to have the ball a lot, have a lot of possession. I am aware that I have to adapt to them.
“Things are going very well for me at Chelsea and that encourages me to keep improving.”
