Spurs manager Mauricio Pochettino has been tinkering with his defence since taking over at White Hart Lane at the end of last season.
Kaboul and Vertonghen, who are facing competition from Federico Fazio and Vlad Chiriches for a starting spot, lined up against Arsenal.
And the Spurs pair impressed as Nacer Chadli and Alex Oxlade-Chamberlain netted for either side in a 1-1 draw at The Emirates.
“Arsenal had a lot of possession but we defended well as a team, we worked very hard,” Vertonghen told Tottenham’s website.
“We have a good understanding, Younes and I, and we fit together well. We dealt with the attackers but as a team we can be happy.”
Vertonghen added that Spurs deserved at least a draw from Saturday’s north London derby.
“It was definitely a deserved point, even though we were 1-0 up quite close to the end, as Arsenal pressed us back. But we stuck together effectively and have come away with a draw,” added Vertonghen.
